Patrick Bamford gave the Blades an early lead before Mark McGuinness headed in a second, with Callum O'Hare adding a late third to secure their place in Monday's second-round draw.
No repeat of a cup shock from last season
Mansfield had won at Bramall Lane in the third round of the FA Cup last season, but hopes of another upset were quickly diminished when Sheffield United struck inside four minutes at One Call Stadium.
Joe Rothwell saw two efforts blocked in quick succession, but when the ball fell back to him for a third attempt, his shot was drifting wide until Bamford turned it in from close range.
Rothwell was then presented with a fine opportunity to double the visitors' advantage midway through the first half, but he fired over after being picked out by Tahith Chong's pull-back.
Mansfield came close to levelling just before half-time, but Sheffield United goalkeeper Michael Cooper made a low save to his left to keep out Frazer Blake-Tracy's volley.
It proved to be the hosts' best opportunity of the night. McGuinness then put the result beyond doubt, powering a header into the net from Sydie Peck's corner. O'Hare completed the scoring late on, finishing a swift counter-attack to put Sheffield United safely into the second round.
